How it Works

Xpert® MTB/RIF delivers results in 2 hours, without making demands on expertise, infrastructure or biosafety measures beyond what can be expected in most peripheral laboratories where direct smear microscopy for TB is routinely performed. The only instrumentation required is Cepheid’s GeneXpert® System, a small-footprint, bench-top device attached to a standard desktop or laptop PC.

Sputum samples (unprocessed or concentrated) obtained for smear microscopy can be assayed individually as they become available. As shown below, the test procedure involves simply adding a reagent to liquefy and inactivate the sputum sample, followed by transferring an aliquot to the cartridge. Neither step demands precise volumes or timing. Hands-on time is less than 2 minutes.

Inside the Cartridge

DNA extraction by ultrasonic cell lysis, real-time PCR, and fluorometric monitoring all take place automatically within the confines of Cepheid’s disposable plastic cartridge; results are sent to the desktop or laptop PC.
